BoneClear: whole-tissue immunolabeling of the intact mouse bones for 3D imaging of neural anatomy and pathology
Dear Editor,Current approaches to analyze cellular structures in the bone tissues have depended mainly on tissue sections.For instance,a recent study visualized different cellular populations in the 300-μm sections of the mouse femurs1.Similarly,the distribution of hematopoietic stem cells could be examined in the half-bone sections2.Notably,such conventional methods obscure specific cellular structures,e.g.,neural innervations,that would be better observed with the intact,unsectioned bones.Moreover,histological sectioning could be challenging for many bone tissues,e.g.,vertebral column,skull and limbs.
