ImmunogenicCharacteristicsofRSVF2ProteinExpressedinE.coliRosetta(DE3)
Respiratorysyncytialvirus(RSV)isamajorcauseoflowerrespiratorytractinfectioninchildrenyoungerthan5yearsold.However,asafeandeffectivevaccinehasnotbeendevelopedsofar,thereforeitisnecessarytoemploydifferentstrategiestoacceleratethedevelopmentofpreventativevaccines.Inthisstudy,thegenederivedfromRSVfusion(F)proteinF2subunitisexpressedinE.coliRosetta(DE3)usingrecombinantglutathioneStransferase(GST)taggedprotein(GSTF2)ofvector.ExpressionofrecombinantproteinisidentifiedbySDSPAGEandWesternBlot.KunmingmiceareimmunizedwithpurifiedF2protein.SeraarecollectedafterthreetimesboosttoassesstheimmuneresponsebyELISA.MiceimmunizedwithrecombinantproteinyieldhightitersofIgG(p<0.05),andtheratioofIgG1/IgG2aisalsosignificantlyhigherthanthatofcontrol.However,thereisnosignificantchangeinsecretaryIgA(sIgA)intherespiratorytractmucosallavagefluidofmice(p>0.01).ResultsshowthattheF2proteinisapotentialvaccinecandidateforRSV,whichinducesTh2typeimmuneresponse.
